Objective To investigate the effect of peroxisome-proliferator-activated receptor-γ coactivator-1α (PGC-1α) on vascular endothelial growth factor (VEGF) expression and tube formation in human vascular endothelial cells. Methods This was an experimental study. Recombinant PGC-1α protein was added to human vascular endothelial cells (HUVEC), and no recombinant PGC-1α protein was added to HUVEC for the control group. After 24 hours of incubation, the two groups of cells were placed into a normoxic (20% O2) or hypoxic (1% O2) environment for another 16 hours. The expression of VEGF mRNA and protein were detected by real-time PCR and ELISA. After 48 hours of incubation, the two groups of cells were plated onto Matrigel-coated plates and cultured under normoxic or hypoxic conditions for another 48 hours. The tube formation of cells was observed by optical microscopy. Experimental data were analyzed by one-way ANOVA. Results VEGF mRNA and protein levels in the cells were significantly increased by recombinant PGC-1α protein both under normoxia and hypoxia conditions compared to the control groups (F=131.67, 65.96, P<0.01). The tube formation ability of cells was enhanced significantly after treatment with recombinant PGC-1α protein compared to the control groups under both normoxia and hypoxia conditions. Conclusion PGC-1α can upregulate the expression of VEGF and promote tube formation in HUVEC.
