Objective To develop a new device which can be used to test contrast sensitivity and color vision effectively and quantitatively in animals, and use it to evaluate contrast sensitivity and color vision in the guinea pig. Methods Experimental study. Based on the spectral sensitivity function of photoreceptors in the guinea pig, three different colors, green, blue and yellow,and yellow-green, at three different levels of contrast sensitivity with 0.6 c/d. For the black-white grating, there were three different levels of contrast sensitivity gratings, 100%, 50%, and 25%, with 3.0 c/d. The interchangeable gratings were attached to the inner side of a drum wall. When keeping the guinea pigs awake, this device was used to evaluate preliminary contrast sensitivity and color vision in the guinea pigs, based on the optokinetic nystagmus (OKN) with a convincing optomotor response phenomena. Kruskal-Wallis test was used to analyze the rate of head tracking movement under different contrast. Results The guinea pig's color vision was demonstrated behaviorally by the OKN response. For 0.6 c/d color gratings with color mixtures of blue-white, green-white, and yellow-green, the guinea pigs exhibited perfect tracking and return movements. At low contrasts, the rate of head tracking movement was 1.06±0.14 (actual contrast, 2.41%), 1.12±0.17 (actual contrast,1.87%), and 1.05±0.15 (actual contrast, 6.50%), respectively. The guinea pigs could still detect black-white gratings with 3.0 c/d when the contrast sensitivity was reduced to 25%. The rate of headtracking movement was 0.74±0.08 at 25% contrast levels, and there was no statistical difference when compared to the high-level contrast gratings by Kruskal-Wallis (x2=2.47, P>0,05). Conclusion The OKN drum could be used to exam color vision in the guinea pig and to quantitatively assess contrast sensitivity acuity. Because of the easy manipulation, it can be used to screen color vision and visual acuity in animals.
